Özet
Konakçı sağlığı üzerinde olumlu etkiler gösteren canlı mikroorganizmalar probiyotik olarak adlandırılırlar. Anne sütü yeni doğmuş bebekler için önemli bir gıda kaynağı olmakla birlikte içeriğinde probiyotik özellik gösteren laktik asit bakterilerini de barındırmaktadır. Çalışmada anne sütlerinden laktik asit bakterileri izole edildikten sonra elde edilen izolatların bazı fenotipik yöntemlerle karakterizasyonları yapılmıştır. Daha sonra izole edilen bakterilerin API 20 Strep kiti ile tür tayinleri yapılmıştır. Yapılan tür tayini sonunda 2 adet Streptococcus salivarius,1 adet Lactococcus lactis subsp. cremoris suşları ile çalışmaya devam edilmiştir. Belirlenen bu 3 suşun bazı starter olma özellikleri incelenmiştir. Tanımlanan bu izolatlar %4 ve %5 lik tuz konsantrasyonlarında gelişme gösterirken, en son 48. saatteki pH ölçümleri incelendiğinde her 3 suşunda asit üretimi pH 5,40 ve pH 6,93 arasında bulunmuştur, S. salivarius (M14-365) suşunun ekzopolisakkarit üretim miktarı en fazla olan suş olarak gözlenirken (8,71 g/L) L. lactis subsp. cremoris (M4-157) ve S. salivarius (M4-11) suşlarının ise EPS üretim miktarları sırasıyla 4,94 g/L ve 5,49 g/L olarak tespit edilmiştir. Tüm suşların nisine karşı dirençli olduğu tespit edilmiştir. Suşların bazı probiyotik olma özellikleri de araştırılmıştır. Üç suşun kanamisin antibiyotiği hariç test edilen diğer antibiyotiklere (ampicillin, gentamicin, kanamycin, erythromycin ve penisilin G) hassas olduğu belirlenmiştir. Ayrıca bu suşların çeşitli insan ve balık patojenleri üzerine antimikrobiyal aktiviteleri araştırılmıştır. Sadece insan kaynaklı patojenlerden olan P. aeruginosa üzerinde S. salivarius suşlarının (M4-11 ve M14-365) antimikrobiyal aktivite gösterdiği belirlenmiştir.
Live microorganisms showing positive effects on host health are called probiotics. Besides milk is an important food source for newborn babies contains lactic acid bacteria showing probiotics characteristics. In this study, Lactic acid bacteria isolated from breast milk. After isolation it has made the characterization of isolates by some phenotypic method. Then the identification of isolated bacteria were done with API 20 Strep system. End of this identification two Streptococcus salivarius and one Lactococcus lactis subsp. cremoris strain are obtained and continued with these strains in this work. Determined these three strains are examined for starter feature. As these isolates have shown growth in salt concentration between 4% and 5%. At last 48 hours, pH concentrations are measured and acid production rate are monitored between pH 5.40 and pH 6.93 for these three strains. Also exopolysaccharide production of these strains was determined and S. salivarius (M14-365) was observed the highest rate (8.71 g/L). EPS production of L. lactis subsp cremoris (M4-157) and S. salivarius (M4-11) strains were determined as 4.94 g/L and 5.49 g/L respectively. It was determined that all strains were resistance against nisin. Some probiotic features of these strains were investigated. And they were tested with antibiotics (ampicillin, gentamicin, kanamycin, erythromycin and penicillin G ) was found to be sensitive to this antibiotics except three strain kanamycin antibiotic. Besides antimicrobial activity on various human and fish pathogens of these strains was investigated. Only S. salivarius strains (M4-11 and M14-365) showed antimicrobial activity against P. aeruginosa pathogen of human.
