Abstract
Patojen mikroorganizmaların sebep olduğu hastalıklar ve antibiyotiklere karşı direnç geliştirmeleri insan sağlığını tehdit etmektedir. Bitkiler eski çağlardan beri çeşitli hastalıkların tedavisinde yaygın olarak kullanılmaktadır. Aronia melanocarpa (Aronya), yüksek besin değeri ile son yıllarda popülerlik kazanan bir meyvedir. Bu tez çalışmasının amacı, Aronya meyvelerinden elde edilen su ekstresinin krem katkılarında ve kaplama materyallerinde doğal katkı maddesi olarak kullanım potansiyelini belirlemektir. İlk olarak ekstrenin gıda, klinik, balık kaynaklı patojenler ve probiyotik adayı anne sütünden izole edilen laktik asit bakterileri (LAB) üzerindeki antimikrobiyal aktivitesi belirlenmiştir. Ekstre patojen mikroorganizmalar üzerinde iyi antimikrobiyal aktivite göstermiştir. Ekstrenin, test edilen konsantrasyonlarında (0.5, 1, 2 mg/disk), Limosilactobacillus fermentum MA-7 üzerinde inhibitör aktiviteye sahip değilken diğer LAB'leri üzerinde düşük antimikrobiyal aktiviteye sahiptir. Daha sonra ekstrenin (0, 50 ve 100 mg/ml), Candida albicans ATCC 10231 ve L. fermentum MA-7 üzerindeki öldürücü konsantrasyonu 0, 24 ve 48. saatlerde makro-seyreltme yöntemi kullanılarak belirlenmiştir. 48. saatte, ekstrenin 100 mg/ml konsantrasyonu, C. albicans ATCC 10231 üzerinde kontrole göre %58.11 ölüm oranına ve 50 mg/ml ekstre konsantrasyonu L. fermentum MA-7 için %135.37 canlılık oranına sahiptir. Ayrıca krem katkılarındaki potansiyel kullanımlarını değerlendirmek için ekstrenin ve ticari krem- ekstre karışımlarının (2.5 - 5 - 10 ml) güneşten koruma faktörü (GKF) belirlenmiştir. Ekstre, 25.77 GKF değerine ve 10 ml konsantrasyondaki krem-ekstre karışımı 13 GKF değerine sahiptir. Son olarak, Aronya su ekstresi ve/veya L. fermentum MA-7 ile geliştirilen antimikrobiyal krem ve yenilebilir film/kaplama materyalinin antimikrobiyal aktivitesi belirlenerek ilaç, kozmetik ve gıda sanayinde potansiyel kullanım olanakları belirlenmiştir. Ekstre ve L. fermentum MA-7'in sinerjik etkisi patojen mikroorganizmalar üzerinde yüksek antimikrobiyal aktiviteye sahiptir. Tez çalışmasında elde edilen sonuçlara göre Aronya meyvesinin çeşitli endüstrilerde birçok sentetik katkı maddesine ve ürünlere alternatif doğal bir ajan olabileceği belirlenmiştir.
Diseases caused by pathogenic microorganisms and their development of resistance to antibiotics threaten human health. Plants have been widely used to treat various diseases since ancient times. Aronia melanocarpa (Aronya) is a fruit that has gained popularity recently with its high nutritional value. The aim of this thesis study is to determine the potential of using water extract obtained from Aronya fruits as a natural additive in cream additives and coating materials. Firstly, the antimicrobial activity of the extract on food, clinical, fish-borne pathogens and probiotic candidate lactic acid bacteria (LAB) isolated from breast milk was determined. The extract showed good antimicrobial activity on pathogenic microorganisms. The extract, at the tested concentrations (0.5, 1, 2 mg/disc), had no inhibitory activity on Limosilactobacillus fermentum MA-7 and low antimicrobial activity on other LABs. Then, the lethal concentration of the extract (0, 50 and 100 mg/ml) on Candida albicans ATCC 10231 and L. fermentum MA-7 was determined using the macro-dilution method at 0, 24 and 48 hours. At 48 Hours, a 100 mg/ml concentration of the extract had a 58.11% mortality rate relative to the control on C. albicans ATCC 10231, and a 50 mg/ml extract concentration had a 135.37% survival rate for L. fermentum MA-7. In addition, the sun protection factor (SPF) of the extract and commercial cream-extract mixtures (2.5 - 5 - 10 ml) was determined to evaluate their potential use in cream additives. The extract has a SPF value of 25.77 and the cream-extract mixture at 10 ml concentration has a SPF value of 13. Finally, the antimicrobial activity of the antimicrobial cream and edible film/coating material developed with Aronya water extract and/or L. fermentum MA-7 was determined and potential uses in the pharmaceutical, cosmetic and food industries were determined. The synergistic effect of the extract and L. fermentum MA-7 has high antimicrobial activity on pathogenic microorganisms. According to the results obtained in the thesis study, it has been determined that Aronya fruit can be a natural alternative to many synthetic additives and products in various industries.
