Zirai mücadelede yaygın kullanılan fungisitlerin gökkuşağı alabalığı (Oncorhynchus mykiss) sperm hücreleri üzerine etkisi

| dc.description.abstract | Bu çalışmada, captan (CPL), mancozeb (MCZ) ve azoxystrobin (AZS) fungisitlerinin gökkuşağı alabalığı (Oncorhynchus mykiss) sperm hücreleri üzerine in vitro etkileri araştırılmıştır. Bu amaçla, farklı derişimlerdeki bu fungisitlere 2 saat boyunca maruz bırakılan spermatozoa örneklerinde motilite parametreleri, lipid peroksidasyon seviyeleri, toplam antioksidan kapasiteleri (TAK), süperoksit dismutaz enzimi (SOD) aktiviteleri, DPPH inhibisyonları ve motilitenin %50 civarına düştüğü örneklerde yağ asidi kompozisyonları tayin edilmiştir. Buna göre, CPL ?2 ?g/L derişimlerde, MCZ ?1 ?g/L derişimlerde ve AZS ?5 ?g/L derişimlerde spermatozoa motilite parametrelerini istatistiki olarak azalttığı saptanmıştır (P<0,05). Benzer derişimlerde, TAK değerleri, SOD aktiviteleri ve DPPH inhibisyon seviyelerinde azalmalar, lipid peroksidasyonunda ise artmalar belirlenmiştir. Değerlendirilen fungisitlerinin spermatozoa motilite değerlerini %50 seviyesine düşürdüğü derişimler CPL için 10 ?g/L, MCZ için 5 ?g/L ve AZS için 200 ?g/L olarak tayin edilmiştir. Bu derişimlerin bulunduğu spermatozoa örneklerinin yağ asidi profilleri incelendiğinde fungisit gruplarının neredeyse tüm yağ asitlerinde değişimlere neden olduğu gözlemlenmiştir. Özelikle çoklu-doymamış yağ asitlerinin miktarlarında önemli azalmalar belirlenmiştir. Sonuç olarak 3 farklı fungisitin, gökkuşağı alabalığı sperm hücreleri kullanılarak farklı toksik etkilere sahip olduğu gösterilmiştir. | |
| dc.description.abstract | In this study, the effects of captan (CPL), mancozeb (MCZ) and azoxystrobin (AZS) fungicides on rainbow trout (Oncorhynchus mykiss) spermatozoa were investigated in vitro. With this aim, spermatozoa samples were incubated with different concentrations of these fungicides for 2 hours, and then motility parameters, lipid peroxidation levels, total antioxidant capacity (TAC), superoxide dismutase (SOD) activities, DPPH inhibitions and fatty acids profiles of spermatozoa samples which are at 50% motility due to the fungicides were determined. Accordingly, ?2 ?g/L of CPL, ?1 ?g/L of MCZ and ?5 ?g/L of AZS caused significantly decreases of spermatozoa motility parameters (P<0,05). At the similar concentrations, decreases in TAK levels, SOD activities and DPPH inhibitions, and increases in lipid peroxidation levels were recorded. CPL at concentration of 10 ?g/L, MCZ at concentration of 5 ?g/L and AZS at concentration of 200 ?g/L reduced spermatozoa motility to around %50 in the relevant samples. When fatty acids profiles of the spermatozoa samples exposure to these concentrations of the fungicides were evaluated, almost all fatty acids profiles have shown differences. Significant reductions particularly in the amount of polyunsaturated fatty acids were observed. Consequently, the different toxic effects of these fungicides have been revealed using with rainbow trout spermatozoa. | |
