    Gossypin protects against renal Ischemia-Reperfusion Injury in rats

    Renal injury occurring as a result of renal ischemia-reperfusion may lead to renal failure or even death. The aim of this study is to investigate possible protective effects of Gossypin on tissue damage occurred due to ischemia-reperfusion in rat kidney tissue. A total of 48 male Wistar albino rats were used in the study. These rats were randomly divided into 6 groups equally (n = 8). The created groups were control (C), sham (S), ischemia-reperfusion (I/R), I/R + DMSO, I/R + 400 mu g/kg gossypin and I/R + 4 mg/kg gossypin. In the rats of sham group, the right nephrectomy was performed. In the rats of other groups rather than sham, the left renal artery was clamped after performing the right nephrectomy. Gossypin was administered intraperitoneally before the reperfusion. 24 h reperfusion was applied to the left renal after 1 h of ischemia. TNF-alpha, IL-1 beta, IL-6 and IL-10 levels were measured with spectrophotometric methods in the kidney tissues after the procedures were completed. Apoptosis and inflammatory pathways were evaluated histopathologically using Caspase 3 and NF-kappa B antibodies. There was a statistically significant decrease in IL-1 beta and IL-6 levels of the gossypin groups compared to the I/R group (P<0.05). As the level of TNF-alpha was decreased in the gossypin administered groups compared to the I/R group although not statistically significant, the level of IL-10 was increased. In the present study, we aimed to show that gossypin in renal I/R model is effective on inflammatory process and apoptosis and that it can be used in routine treatment to decrease the damage in all reasons that may cause I/R. In addition, this study can shed light on the studies to be done in this field in the future.

    Protective Effects of Baicalein and Bergenin Against Gentamicin-Induced Hepatic and Renal Injuries in Rats: An Immunohistochemical and Biochemical Study

    Drug-induced organ toxicity is a significant health concern, with gentamicin known for its effective antibacterial properties but also severe side effects, particularly cytotoxicity in liver and kidney tissues. This current study observed the preventive role of baicalein and bergenin against hepatic and renal injuries caused by gentamicin in rats. Methods: Thirty-two male Sprague Dawley rats were divided into four groups, namely, control, gentamicin (gentamicin 80 mg/kg/day), baicalein (gentamicin 80 mg/kg/day + baicalein 100 mg/kg/day) and bergenin (gentamicin 80 mg/kg/day + bergenin 100 mg/kg/day). Hepatotoxicity and nephrotoxicity were induced by giving gentamicin (80 mg/kg/day). We evaluated the biochemical markers, including alkaline phosphatase (ALP), urea, alanine transaminase (ALT), creatinine and aspartate transaminase (AST) levels, antioxidant enzymes, oxidative stress parameters and histopathological and immunohistochemical changes. Results: Gentamicin increased oxidative stress parameters and decreased antioxidant activity. The treatment with baicalein and bergenin significantly restored these markers. Conclusions: Baicalein and bergenin significantly mitigated gentamicin-induced hepatic and renal toxicity by restoring biochemical markers, reducing oxidative stress and enhancing antioxidant enzyme activity. Histopathological and immunohistochemical analyses confirmed the protective effects of both compounds against organ damage. No statistically significant differences were observed between the two drugs for these parameters. These results suggest their potential as therapeutic agents to prevent gentamicin-induced organ toxicity.

    The Protective Role of Syringic Acid on Ovarian Injury Created by Ischemia Reperfusion

    Syringic acid (SA), a natural phenolic substance, possesses anti-inflammatory and antioxidant properties. We investigated the likely preventive effects of SA on organ damage to the reproductive system caused by bilateral ovarian ischemia -reperfusion (I/R). Materials and Methods: 32 female Sprague-Dawley rats in total were used; they were split into four groups: a sham group for co ntrol (group I), I/R group (group II), I/R group given 10 mg/kg SA (group III), and an I/R group given 50 mg/kg SA (group IV). Grou p I underwent only an abdominal incision without I/R. In groups II, III, and IV, I/R procedures were conducted for 3 hours. S A was given intraperitoneally to groups III and IV for 30 minutes before reperfusion at 10 mg/kg and 50 mg/kg, respectively. The ovarian tissues were surgically removed for analysis. Results: TNF-?, MDA, IL-1?, MPO, TOS and OSI levels in group II were found to be significantly higher than in group I. Additionally, a decrease was detected in TAS and SOD values in group II. In contrast, SA administration led to significant increases in the level of antioxidant enzymes in both groups III and IV. Also, in the SA-given groups, significant decreases were detected in OSI and TOS levels, along with a decrease in MPO, MDA, TNF-?, and IL-1? levels. Conclusion: It has been demonstrated that SA has a protective effect against organ damage caused by bilateral ovarian I/R in the reproductive system. This is the first study to show that SA can reduce ovarian damage due to I/R injury.
