Öğe Antibacterial, antioxidant, and angiotensin-I converting enzyme inhibition effects of margotia gummifera extracts rich in phenolic compounds(2022) Bouchekrit, Moufida; Laouer, Hocine; Çakmak, Yavuz Selim; Hajji, Mohamed; Boutellaa, Saber; Nasri, Moncef; Akkal, SalahAromatic and medicinal plants have been used to substitute synthetic drugs with natural bioactive products. Objective: The current investigation was conducted to evaluate phenol and flavonoid contents, antibacterial, antioxidant, and anti-hypertensive potentials of three extracts of Margotia gummifera aerial parts. Methods: The antibacterial effect was assessed using the wells agar diffusion method against 11 strains. The antioxidant effect was evaluated using different methods, 2, 2-diphenyl-1-picrylhy-drazyl, total antioxidant capacity, ferric reducing ability power, cupric reducing antioxidant capaci-ty, and ?-carotene/linoleic acid bleaching assay. The anti-hypertensive activity was performed using hippuryl-histidyl-leucine substrates. Results: The highest yield, i.e., 19.801%, was obtained by the hydro-methanolic extract. However, ethyl acetate extract seemed to be rich in phenolic and flavonoid compounds compared to other ex-tracts, i.e., 822.14±69.10 mg GAE g-1 ext. and 117.28±1.80 mg RE g-1 ext., respectively. The high free radical scavenging activity was mentioned in ethyl acetate extract with IC50 of 48.820±1.25 ?g/ml. The same extract showed better antioxidant activity in all tested procedures. In antibacterial activity, the hydro-methanolic extract exhibited moderate effect against all tested bacteria except Salmonella enterica and Enterococcus faecalis which gave 18±2.0 and 16±1.0 mm of inhibition, whereas it seemed to be inactive towards two Gram-negative bacteria. Concerning anti-hyperten-sive activity, all extracts can inhibit the Angiotensin-I Converting Enzyme, but the potential effect was given by hydro-methanolic and aqueous extracts, 97.75% and 96.65%, respectively. Conclusion: It can be stated that the bioactive compounds of Margotia gummifera extracts had different biological activities, which confirm their therapeutic uses in traditional medicine.Öğe Helosciadium crassipes Koch (Apiaceae) extracts as natural sunscreen and preservative additives(Horticulture and Forestry Society from Transylvania, 2023) Makhloufi, El-Hani; Lefahal, Mostefa; Ayad, Radia; Çakmak, Yavuz Selim; Nieto, Gema; Akkal, SalahRecently, ingredients derived from natural sources have gained increasing interest in the cosmetic field due to their low toxicity. Therefore, this study was designed to explore the cosmetic potential of Helosciadium crassipes Koch (Apiaceae) species growing in Algeria as a promising natural preservative and broad-spectrum UV protection additive. In our study, the in-vitro antioxidant effect was assessed via DPPH radical scavenging and total antioxidant capacity by phosphomolybdenum method (TAC), while the protection against UVB radiation was evaluated according to the sun protection factor (SPF) by using UV spectroscopic technic at wavelengths ranging from 290 to 320 nm and Mansur’s equation, for the photoprotective effect against long-wavelength UVA, UVA/UVB and critical wavelength (?c) parameters were evaluated. The outcomes showed that among the tested extracts, the methanolic extract (MeOH) contains high levels of phenolics and flavonoids, and possesses a significant antioxidant effect, particularly in DPPH radical scavenging assay. Similarly, this last one exhibited high photoprotective activity in UVB and UVA ranges. The gathered results reveal the possibility of using this extract as a good natural additive to be incorporated into cosmetic formulations as a broad-spectrum UV protection candidate and as a preservative agentÖğe In vitro antioxidant and antimicrobial activities of aerial parts of Algerian Jurinea humilis DC (Asteraceae)(Pharmacotherapy Group, 2017) Ayad, Radia; Çakmak, Yavuz S.; Özüsağlam, Meltem A.; Medjroubi, Kamel; Akkal, SalahPurpose: To assess the polyphenolic composition of various solvent extracts of Jurinea humilis DC. as well as their in vitro biological effects. Methods: The crude extracts of the aerial parts of Jurinea humilis were obtained by maceration method with dichloromethane (DCM), methanol (MeOH) and ethyl acetate (EtoAC), separately. Folin-Ciocalteu and aluminum chloride procedures were used to quantify the total phenolic and flavonoid contents, respectively, while antioxidant properties were determined by two methods: phosphomolybdenum and DPPH assays. Antimicrobial activity was also evaluated by disc diffusion method. Results: The ethyl acetate extract exhibited the highest amount of total phenolic content (TPC, 169.14 +/- 7.22 mg gallic acid equivalent/g sample) and total flavoniod contents (TFC, 104.91 +/- 0.22 mg rutin equivalent/g sample). The extract (EtoAC) also showed the strongest antioxidant activity of the three extracts, especially DPPH assay (IC50 = 0.16 +/- 0.00 mg/mL), as well as demonstrated the highest activity against the pathogenic strains tested. Conclusion: The results suggest that J. humilis DC. is rich in phenolic and flavonoid contents which may be responsible for good antioxidant and antimicrobial properties. Thus, J. humilis may be a promising source of new antioxidant agents and pharmaceuticals.
